| I. Background | |
| IL-1 家族包括 11 種細(xì)胞因子，7 種具有激動劑活性的配體（IL-1α，IL-1β，IL-18，IL- 33，IL-36α，IL-36β，IL-36γ）和 4 種具有拮抗活性的成員[IL-1 受體拮抗劑（IL-1Rα）， IL-36Ra，IL-37，IL-38]。IL-1α，IL-1β，IL-18，IL-33，IL-36α，IL-36β，IL-36γ觸發(fā) MPKK 和 NF-κB 導(dǎo)致炎癥反應(yīng)。 與 IL-1 家族的其他細(xì)胞因子成員類似，IL-36 激動劑（IL-36α、β、γ）與由 IL-36R 和 IL-1R 輔助蛋白（IL-1RAcP）組成的異二聚體受體結(jié)合。IL-36R 由許多不同的細(xì)胞類型表達(dá)，包括角質(zhì)形成細(xì)胞、肺成纖維細(xì)胞和上皮細(xì)胞、內(nèi)皮細(xì)胞以及各種免疫細(xì)胞。IL-36R 在人 M0 和 M2 巨噬細(xì)胞中也高表達(dá)，但在 M1 巨噬細(xì)胞中不高表達(dá)。IL-36Rα 和 IL-38 對 IL-36 信號通路具有負(fù)向調(diào)節(jié)作用。IL-36Rα通過與 IL-36R 競爭性結(jié)合、抑制 IL-36 激動劑 識別和 IL-1RAcP 募集來表現(xiàn)出拮抗功能，從而抑制該家族的激動成員對受體的激活。除 了阻斷 IL-36R 介導(dǎo)的 MAP 激酶和 NF-κB 通路的激活外，IL-36Ra 也可以阻止 Th17 細(xì)胞因 子的表達(dá)。 | |
